Extracellular and Metabolic Factors Affecting the Efflux and Influx of Erythrocyte Water

چکیده

Water exchanges between rabbit erythrocytes and extracellular solutions equidistant from intracellular osmolarity were studied by freezing point depression techniques. Water efflux was always less than water influx and both were hypotonic to the intracellular and extracellular fluids. The magnitudes of these water exchanges were not dependent on the presence of extracellular cation. Stimulation of oxygen uptake by the addition of glucose and methylene blue increased water influx and, possibly, decreased water efflux. This could not be accounted for by accumulation of osmotically active intracellular metabolic products. O(2) uptake was markedly decreased during cellular dehydration, was slightly decreased during cellular overhydration, and was maximum at the water content of erythrocytes when suspended in a medium isotonic with plasma.

Evaluation of magnesium fluxes in rat erythrocytes using a stable isotope of magnesium.

The mechanisms that maintain intracellular Mg concentration at physiologic levels are not fully understood. In this work, we described for the first time, a new method using 25Mg stable isotopes, that allows simultaneous determination of Mg2+ efflux and Mg2+ influx in non-loaded cells at physiological levels of extracellular Mg. Erythrocytes from rats were suspended as a 10% suspension in NaCl ...

Physiological Responses of the Crayfish Pacifastacus Leniusculus to Environmental Hyperoxia I. Extracellular Acid-base and Electrolyte Status and Transbranchial Exchange

Extracellular acid-base and ionic status, and transbranchial exchange of acidic equivalents and electrolytes, were monitored in freshwater crayfish (Pacifastacus leniusculus) during control normoxia (POz = 148 mmHg; 1 mmHg = 133-3 Pa), 72 h of hyperoxia (PO2 = 500 mmHg) and 24 h of recovery. An initial (3 h) respiratory acidosis of 0-2 pH units was completely compensated within 48 h by a 50% in...

Lactate transport by rainbow trout white muscle: kinetic characteristics and sensitivity to inhibitors.

This study used an isolated-perfused tail-trunk preparation of rainbow trout to examine the uptake and release of lactate (Lac) and metabolic protons (delta H+M) in resting and exercised fish white muscle. In exercised muscle, L(+)-Lac efflux was inhibited (approximately 40%) by 5 mM alpha-cyano-4-hydroxycinnamate (CIN), but not by 0.5 mM 4-acetamido-4'-isothiocyanostilbene-2,2'-disulfonic acid...

Transport of monosaccharides. I. Asymmetry in the human erythrocyte mechanism.

Transport of D-glucose across human erythrocyte membranes occurs via a facilitated diffusion process which demonstrates influx-efflux asymmetry. The mechanism of the asymmetry has been studied by estimating unidirectional fluxes in the presence or absence of trans equilibrium hexose. In the absence of transhexose, the half-saturation constant for efflux at 15 degrees C was approximately 10 mM a...

The effects of chronic hypokalaemia, hyponatraemia, and acid-base alterations on erythrocyte sodium transport.

1. Erythrocyte sodium concentration and fluxes were measured in patients with acid-base disturbances, hypokalaemia and hyponatraemia. Results were similar to those obtained with normal erythrocytes exposed to artificial in vitro alterations. 2. Erythrocyte sodium content and influx varied directly with extracellular bicarbonate which appeared to influence membrane permeability. 3. Hypokalaemia ...
